Hi everyone- I've made small batches with success, now I'm looking to make larger batches for myself so I only have to make them like once a year. I did some research and put together a quick guide to make it easy for me. I took some info from searching this forum combined with my own knowledge. I hoping to get some insight to perfect this guide, which can bring value not only to myself but also everyone here. What am I missing or could be improved on ? Also put some questions within the steps of things I'm unsure of. Any advice would be greatly appretiated and I'm willing to offer any help I can as well!


Step 1:

Sterilize Media bottle- Wash with hot water & 10% bleach- Then wash with hot water and dish soap- Dry it oven at 400 for 30minutes or until completely dry

Sterilize Beaker- Wash with hot water & 10% bleach- Then wash with hot water and dish soap- Dry it oven at 400 for 30minutes or until completely dry

Sterilize 10ml vials – When using many vials to fill, washing them individually doesn’t seem like the most effective way. What is the best way to sterilize 10ml open vials?

Step 2: Weigh powder using digital scale

Step 3: Add powder, BA, BB and grapeseed oil into the Beaker.

BA- Benzyl Alcohol, Anhydrous, 99.9% Pure, USP 500mL

BB- Buy Benzyl Benzoate USP Grade, Ultra Pure, 500 mL

Oil- Buy Grapeseed Oil by Vale of Eden, 1 Gallon (Approx. 4 Liters)

Step 4: Place Beaker on Hot plate/Magnetic Stirrer

Heat to 250f - Keep an eye on the powder and watch as it dissolves until it gets close to clear. A clear solution means that the mixture is now melted.

All this contributes to dissolving the powder entirely into the solvents. Move to the next step when no powder is visible.

Step 5: Filtering

Filter while solution is warm (but not hot) Around what temp would you recommend for this stage of filtration ?

Place Whatman filter on top of sterilized media bottle

Pour from beaker onto Whatman filter

Connect pump to Whatman filter

Step 6: Bottling and crimping

Once filtration is complete- Remove Filter & Place bottle top dispenser on top of media bottle

Pour 10ml into each 10ml vial from media bottle with bottle top dispenser

Place rubber stoppers on top of each vial

Place flip off caps on top of each vial- crimp with crimper

Confused how you can use bleach, wouldnt rubbing alcohol work better?

PSA - you do not need to heat or pressure sterilize your lab ware it is completely unnecessary.

I shake it up until it’s mixed. I then screw the cap off and stick a reused syringe and pull out 3 ml. I filter the contents of that syringe directly into and other syringe. Then I screw a needle on and then I shoot up. Yes it is that simple.

you want to rinse with distilled water a couple times after soap and water otherwise will have soap residue left behind. SOP is 3 rinses in the lab.

I repeat those water bottles that filter sh*t water with AIDS and ebola into drinkable water in sh*thole countries use filtration. Not only can you drink that water but you can inject it ffs.

This is just not accurate. Eye drops are formulated for eyes. They can be used in the ears, whereas, ear drops cannot be used in the eyes. Why? Because for example they do not put benzyl alcohol in eye drops. Because they go in your eyes. Poor analogy.

And 2nd analogy is useless as well. Dry vials are not in anyway remotely the same as water pipes. This makes no sense. If you want to disinfect the vial with IPA before you poke it then that is more effective than using an autoclave at any point in this process.

But you don’t even need to keep your finished product in a vial. I keep mine in a Nalgene bottle. …

The Ebola virus has a diameter of 80nm, and the common 0.22 um filter can only filter out 220nm residuals. If you really need to filter water in that kind of condition, the 0.22 um filter will certainly be clogged in a sec. Then you need a multi-stage filter to make sure you don't clog the 0.22 um filter, and add more steps to remove heavy metal ions because they can't be filtered out, such as dual ion-exchanged units (one for positive ions and one for negative ions), add UV lights to kill the virus and distill them to make sure other insoluble stuff is removed. Then we have a modern water purification facility, which shows why 0.22 um isn't sufficient to meet our standards.

PSA - you do not need to heat or pressure sterilize your lab ware it is completely unnecessary.

and ditch the hot plate and stirrer If you need to heat anything to 250 F then it’s too concentrated.

You know how I do it? I get a plastic bottle that I have reused 10 times and wash it out with dish soap and water and dry it. I pour the oil, powder, BA, BB, all in to the bottle. I screw the cap on. I shake it up until it’s mixed. I then screw the cap off and stick a reused syringe and pull out 3 ml. I filter the contents of that syringe directly into and other syringe. Then I screw a needle on and then I shoot up. Yes it is that simple.

I'm not the kind of guy to argue or debate, but I'll just ask a question. Rather I'll make a statement. Every compound has it's own melting point for a reason. In order to reach that melting point unless you live in the Sahara Desert and you want to use a magnifying glass to bounce the rays off the Sun omto your Raws you're going to need heat. Preferably a hot plate with a PTFE magnetic stir rod. But then again what do I know..

water filtration does not = .22 filter.. nor does filtering the water clean the shit bucket it came out of. to use your analogy.

again, will point to a GMP FDA facility recently having bacteria in eyedrops that is killing people with regulated packaging a process etc.

not sterile viles may have god knows what on them from manufacturing putting filtered oil in said vile does not clean that. also will not clean the manufacturing oils etc etc.

Think about it like washing your hands, you never will get all germs off your hands, but yet its a good idea because it gets ALOT off and therefore will get sick far less.

1 last analogy, think of it like Mexican water, Mexican water treatment facility clean there water perfectly fine, but what makes people sick is it running through DIRTY pipes(vials in this analogy) that pick up god knows what. and while yes u will get small amounts in nose or mouth taking a shower drinking a glass may cause disease. the more corners u cut its like adding more length to the Mexican pipes thereby increasing risk. clear as mud?
