Giant Semaglutide Thread (and other GLP-1 / GIP agonists)

Increased my reta dose this week, at 18mg/week now.
That stupid feeling on the skin is back!
Appetite suppression is back too, haven't felt that..in a very long time.

The annoying skin feeling seems to be completely gone. I have been using ARA290 for about a week now and I think it's helping but it might just be placebo, probably will go up to 20mg next month.
 
Big event next week, after a year and four months on tirz, I’m finally switching over to reta, starting at 2 mg. For those who’ve made the jump, did you actually feel that burst of energy everyone keeps hyping up, or is it just another one of those “depends on the person” stories?

I feel lethargic on tirz. On reta I feel stimulated a few minutes after taking it and don't have a justifiable reason for coffee
 
1 mg tirz, 1 mgs sema, is highest Im going this year, and it seems plenty

Three weeks untill competition date and dont think Ill take anymore of this then about once per week. When Im not able to get calories in ill smoke a bit of cannabis, and this helps eating more

Not recommending cannabis but if hunger just isnt there, or for a carb up, or carb and fat load , it seems to definately make it easier to get down food

What's your reasoning for 1mg Tirz out of curiosity?

The potency is roughly 1/6th that of Sema (15mg vs 2.5mg max RX doses)

So 1mg Tirz = approx 0.15mg Sema

Not sure 0.15mg Sema would do anything lol

I only have 5 mgs left , and not sure if I’ll advance to nationals this year if I win this next competition . Nationals is mid December so glps may run out and these competition s costs, esp if not sponsored and pay for them

From an economic standpoint sema wins and more than likely the only glp I’ll spend

Yeah probably not. I mentioned it in this thread once before but I paid 190$ for 10 mgs tirz from MA research last year This year 40$. I got 5 mgs sema for 20$ recently

I only have 5 mgs left , and not sure if I’ll advance to nationals this year if I win this next competition . Nationals is mid December so glps may run out and these competition s costs, esp if not sponsored and pay for them

Sending someone a few bucks worth of GLP-1's if it significantly impacts their BB'ing career is a no-brainer.

So, basically, people experience less metabolic adaptation with Tirz and Sema, and even less with Reta, compared to traditional dieting without GLPs?
When they gave people tirz and stuck them in an indirect calorimeter, they found that there was no reduction in metabolic adaptation compared to traditional dieting.

A similar clinical trial is currently in progress for reta.

My guess is that it depends on how jacked up your liver is. Reta specifically shreds fatty liver and in doing so dumps a bunch of excess energy and ketones. If you’ve got a lot of fatty liver to burn off you can get a good energy/ketone bump going for awhile. If you’re healthier then that’s not going to happen for you.

Also, for Reta, would you suggest starting the titration from the initial dose again, or is it fine to begin at 1 or 2 mg?"

I’d start very low .5mg few weeks then go up another .5mg -1mg
 
It's alot weaker according to that table, and given how the dosing is lesser too, I'd expect lesser sides too.
In higher doses, mazdutide is extremely effective. Granted that both studies I cite are small. However, they achieved their weight loss with patients on the high doses for extremely short time periods. Ji, et al. (2022). Safety and efficacy of a GLP-1 and glucagon receptor dual agonist mazdutide (IBI362) 9 mg and 10 mg in Chinese adults with overweight or obesity - a randomised, placebo-controlled, multiple-ascending-dose phase 1b trial. EClinicalMedicine 54, 101691; Bhattachar, et al. (pre-publication, 2025-08-20). Mazdutide reduces body weight in adults with overweight or obesity - A high-dose Phase 1 trial. Diabetes Obesity & Metabolism, doi.org/10.1111/dom.70040.

When looking at studies of mazdutide, it's important to distinguish between studies conducted in China and those in the US. The populations in China tend to have far lower BMIs to begin with. As a result, they are likely to lose less weight.
