Media Bottle to Vials

Evening all,

I'm about to embark on my first bottle top filter brew, however I've encountered a slight road block.

What's the best way of transferring from the media bottle to my open vials? I've been reading a few options but cant seem to knuckle down a process....

Any help would be great, Cheers

Depends on how paranoid you are about the risk of contamination (and how many vials you are prepping).

"Most risk":
Pump from media bottles with vacuum pump through sterile hose into the open top of your sterilized vials and then cap. (risk is still pretty bloody minimal unless you are doing this in a chicken coop).

Least risk:
Do this (the "Most risk" method) in under a sterile sealed workspace (often unattainable).

Moderate risk:
Cap your vials (I just buy precapped and sterilized vials because fuck all that) and just fill the vials the old fashioned way with a 10 or 20 mL syringe and vented needles (or just shove a small guage needle in alongside the filler needle. This method sucks if you are trying to do like 100+ vials though.
 
I get have the funnel trennedoutlunatic is talking about and sterilizing it and setting it up and gauging how much oil is going in the vials is a pain the the ass.

Now i get a 10ml syringe, and these blunt tip drawing needles

I pull liquid in a little bit above the 10ml mark, to the same spot for every vial. Super simple and easy. Throw the syringe away, and when i go through all the drawing needles, i wash them with soap and water, rinse with iso, ans put in oven at 450F for 2 hours. They are all metal so they can be cleaned easily

 
Borosilicate separatory funnel with PTFE stopcorck.
Disassemble stopcorck.
Dry heat sterilization for the funnel.
Steam or chemical sterilization for the stopcorck.
Laboratory stand.

I had a 5L or 6L sep funnel many moons back as well as some smaller ones, also a couple 10,000ml flasks and a 5L pressure equalized addition funnel amongst my assorted glassware, lol.
 
Media bottles with a 33mm neck topped with butyl septa caps changed everything for me. Filter into the bottle, throw the cap on and draw straight from the bottle as if it were a large vial.
I wouldn’t like that, way too many pokes on the rubber if it’s large volume.

Sick. I just made a post basically asking if this was possible - do you mind checking it out?

Also, what size media bottles do you use? Is 500ml overkill (overkill with regards to the septa/stopper's integrity)?
 
You can use what ever size you want you just need to make sure the neck size is 33mm not 45mm, or it will cost alot more.


So if I was to pierce a large 500ml media bottle however many times it takes to empty, the septa would still be in decent condition? (I'd still toss it and replace it with a new one once the bottle was finished).

That's a good idea. I definitely hadn't considered the cost, but I was planning to use 45mm mainly because that would attach to most bottle top filters. Also, please could you check out my post?

I'd appreciate your insights because the stoppers that I've been looking at are different to what you're recommending.
 
No I would not use it like your saying it will break up over time. The main reason for using is keeping everything sterile while transferring from media bottle to vials. Both stoppers will work but the ones I linked alot more affordable. Or you could do what I'm about Todo and use the caps that come with the gl45. Drill out the center and use these ptfe silicone septa that you insert in the cap.

I was looking at those septas too actually - I've read that the silicone holds up better over time (in response to frequent pinning). How do you bind/attach the septas to the caps though? Do they just stick there by themselves?

Sounds like you're doing exactly what I'm doing though - just with a DIY approach instead of buying the pre-made GlL45 open aperture caps.
 
Yepper... Those septas are made for these exact caps nothing else. So I'm guessing they fit snug underneath the cap and the pressure when screwed on would keep it from leaking. Will be finding out shortly. Actually made a post about it for anyone that's used before but no hits. Should work as stated above though.

We made similar posts at similar times... How odd...

Anyways, if you don't fancy drilling the holes yourself, you could take a look at these ones that I linked in my post - they might be suitable if you can find septas with the corresponding diameter:
