I just backfill a 60ml syringe.
Directly into sealed sterile vials.
Air purifying in room for 6 hours.
2x4 across 2 stands, at stomach level.
2x4 has hole drilled that the 60ml syringe fits through.
Wipe down 2x4.
Wipe off tops of sealed sterile vials.
Place 27G into first sterile vial for venting.
Place 60ml syringe through hole in 2x4.
Place 20G needle on end of syringe.
Remove plunger (this stucks air through syringe of course). Don't let rubber of plunger touch anything.
Immediately place 20G into sealed sterile vented vial.
Open media bottle and pour directly into 60ml syringe.
Immediately recap media bottle.
Then Immediately put plunger back into 60ml syringe and there will be small air pocket on top of gear in syringe.
The gear will Immediately start flowing into syringe.
When gear gets to almost fully in vial, pull plunger back slightly to relive pressure. Otherwise when you remove vial gear will flow from 20G and waste.
Remove vent 27G first and vent next vial, have new vented vial in hand.
Remove full vial and Immediately Place new vented vial on end of 20G.
Restart pressure and fill new vial.
Once syringe empty, don't push the air pocket into final vial.
If the last vial was not full, leave vent in and remove vial from 20G.
Quickly pull back plunger, and Place vial back on 20G. You don't want to remove plunger while 20G is in vial.
Refill syringe and repeat.
There will be some drips once syringe is empty and you remove vial to remove plunger. Then Quickly placed vial back on end of 20G before refilling syringe.

I've never had anything floating in my vial. I've never had an infection.
