The isolation of melatonin was first reported in 1958. Since the demonstration that pineal melatonin synthesis reflects both daily and seasonal time, melatonin has become a key element of chronobiology research.

In mammals, pineal melatonin is essential for transducing day-length information into seasonal physiological responses. Due to its lipophilic nature, melatonin is able to cross the placenta and is believed to regulate multiple aspects of perinatal physiology. The endogenous daily melatonin rhythm is also likely to play a role in the maintenance of synchrony between circadian clocks throughout the adult body.

Pharmacological doses of melatonin are effective in resetting circadian rhythms if taken at an appropriate time of day, and can acutely regulate factors such as body temperature and alertness, especially when taken during the biological daytime.

Despite the extensive literature on melatonin physiology, some key questions remain unanswered. In particular, the amplitude of melatonin rhythms has been recently associated with diseases such as type 2 diabetes mellitus but understanding of the physiological significance of melatonin rhythm amplitude remains poorly understood.
 
I find melatonin supplementation interesting. They are using it in drug/alcohol rehab situations too these days. I recall when it hit the market in the 90's and just .5mgs would whack your ass. But then I was in early 20's and maybe I had more. But the thought of a 3mg dose blew my mind when I started seeing it on shelves..

- When early 20's, I could take it at 4 in the afternoon and go from wide awake to crashed... I know cause a buddy of mine, warned me, gave me one and chuckled...

- Strangely, the rare occasions I took it when young, I recall waking up in the middle of the night with all the blood vessels blown up on one hand, foot, or something weird. Perhaps just sleeping on it that dead to world.?? But it seems to have an effect of sleeping BP for sure..

- Its about as tolerating as Xanax, WHICH MEANS FAST. I know NOW why they have larger doses. I know a guy that was recently taking like 8mgs on top of another sleep aid..

- It does hang around the next day, but not only, I could never time right so it def messes with natural sleep rhythm..

- I catch some rays today a couple times a week, and get knocked out for a LONG TIME. Something is out of whack for me. I got to kick it by a pool for the whole summer of 2008 and was up all day and slept great every night getting up early.

- How do other drugs and alcohol affect?? Im pretty sure alcohol intensifies..

- I have never taken long term and had to opp to determine how it effects diet, energy, exercise, and BMI... I have a suspicion it would be playing with fire... But an extinguishable fire...
 
[OA] Melatonin Regulates the Synthesis of Steroid Hormones on Male Reproduction

Melatonin is a ubiquitous molecule and exhibits different effects in long-day and short-day breeding animals. Testosterone, the main resource of androgens in the testis, is produced by Leydig cells but regulated mainly by cytokine secreted by Sertoli cells.

Melatonin acts as a local modulator of the endocrine activity in Leydig cells. In Sertoli cells, melatonin influences cellular proliferation and energy metabolism and, consequently, can regulate steroidogenesis. These suggest melatonin as a key player in the regulation of steroidogenesis.

However, the melatonin-induced regulation of steroid hormones may differ among species, and the literature data indicate that melatonin has important effects on steroidogenesis and male reproduction.

I use it a few times a week. Weird thing about it is. Every time I use it if I wake up in middle of the night I see spiders on my wall that aren't there. Even at 2mg. No idea why.
 
I tried it just recently.
5mg didn't really do that much, hard to say.
But then I took 10mg and I slept like a rock and then also slept threw most of the next day haha.
