Semaglutide preparation

So i ordered and received semaglutide raws thinking I can make an oral preparation.
How do I make it as an injectible as it should be? I have bac water or nacl water and i can add BA..
Mix powder with nacl water, put in vial, heat up to sterilize and add BA?
I have sterile vials none already opened and uncrimped.
 
Not one suggestion? Can i just brew it in oil? At like 20mg/ml and use slin pin?
Semaglutide is a peptide and soluble in water. You would reconstitute it with sterile water or bac water, like hGH.

Keep refrigerated and out of light, as temperatures over 49F and light exposure increase rate of degradation.

Ozempic uses a phosphate buffer and a stabilizer to help prevent peptide degradation, but without intimate knowledge of chemistry, it might be a bit difficult to achieve the same formula as ozempic, though arguably it would increase efficacy and/or stability.

No reason to use a solvent like BB or oil.

Yes i read about the phosphate stabilizer and without it your stomach just destroys the peptide if ingested orally.
I know how to reconstitute my question is what best procedure if i have sealed vials and bac water? Can i mix in a third mediun then sterilise with syringe filter on way into sealed vial? Will that destroy the peptide?

Tough question. I have bought vials from Qingdao with 2mg and simply reconstituted with bac water.
Your issue is that you dont have them in sterile vials already but as a raw powder in some kind of bag i assume..
mixing it with BA should get rid of most of bacteria and since you are injecting subq it might be less of an issue. i would probably sterilize the vials, mix the sema with BA and push it through a 22µ filter in the vials..

The vials are already sterile i have 10ml and 20ml theyre sealed my only concern is mixing safely.
Maybe if i just mix directly in a syringe then run it through the .22 filter into a new vial.
Peptide molecules are small enough no? But i will still lose an amount and accuracy.
Also scale accuracy, i need to make a large enough amount to use like 50mg, but then ill have enough solution for half a year if taking 2mg a week and it degrades before then.
I cant even reliably scale out 50mg ill have to do 100 or 200mg probably and throw out unused, or provide it.
Edit shit dose is even smaller from 0.25mg a week titratrated up slowly over 15 weeks to 2mg.

To expand on this, here is the information from the patent:

The solution used to reconstitute semaglutide contains:

-0-0.01%w/w phenol
-pH between 7.0-7.8
-isotonic agent (14mg-30mg/mL of composotion)
-buffer (sodium dihydrogen phosphate, disodium hydrogen phosphate, and sodium phosphate @ .01-50mM of compositon)

Unless otherwise noted, compositions of semaglutide were prepared by dissolving buffer (e.g. disodiumhydrogenphosphate dihydrate), isotonic agent (e.g. propylene glycol) and optionally preservative (phenol) in water. Semaglutide was dissolved therein, pH was adjusted to 7.4 using sodium hydroxide and/or hydrochloric acid, and the composition was finally sterilised by filtration through a 0.22 μιη sterile filter.

This all revolves around stability of the molecule. How or why this would effect the results once in the body is beyond me. The only thing I know is the propensity for semaglutide to bind to albumin. Perhaps the pH being 7.4 increases the potential for the peptide to bind to albumin, since the highest affinity binding site is mostly pH dependent - and the big if being: if I understand this correctly.
