transfer from media bottle to vial

hi all

I am going from simple old school brewing ie. sterile vial/caulk gun to trying bottle top option,
i have an autoclave so I can sterilize the media bottle and vials.

how is everyone transferring from media bottle into open-top vials? thought of a few ways

1. bottle top dispenser - bit pricey but most sterile
2. use of septa then perhaps 50ml syringe and then remove needle and squirt into the open vials the crimp 10ml at a time
3. transfer into something sterile like a sterile packed semination cup then use a 10 syringe to squirt into the vials (not too sterile unless done in a laminar flow hood)
4. using a 10ml stripette and pipette boy - again sterility may be an issue

An easy way to accomplish this is to get 10ml pipettes and then punch a hole in a septa the size of the pipette. You will have minimal exposure time while dispensing into the vial. The media bottle with the punched septa will hold the pipette upright while you stopper the filled vial and get the next vial ready

What a great idea mate thanks ! I could probably use a 18g to make a hole then push the pipette through to make bigger

The pipette is way too large for pushing through an 18g hole. You either have to cut the hole or use a punch to punch it out

make sure it’s a model that is autoclavable. Get one with max dispensing of 10ml so probably it would be 2-10ml adjustable. I say get a maximum 10 because unless you spend quite a bit of money, the dispensers like to dispense an unoticable amount more on every pump. Over 50 vials you will be able to see the first ones have slightly less oil than the last ones. If you use a maximum 10ml version the amount of variation seems to be a lot less. Don’t know why that is but I imagine it has something to do with if it’s maxed out there isn’t much more room for it to fill the reservoir up with extra oil as in minutely adjusts itself every pump. I got one for 200$. Seems to be the cheapest one I could find that was autoclavable that also had decent reviews. There are cheaper versions but they are said to be garbage toys.

Once your done filtering you just unscrew bottle top filter and screw on a sterilized dispenser, it has an telescoping tube to help fit different sized media bottles, it also comes with 5 different adapters to make it fit a variety of sized openings.

How do you clean it in between uses? Obviously you autoclave it, but what about cleaning out the oil from the previous batch?
