Giant Semaglutide Thread (and other GLP-1 / GIP agonists)

Upped my perfect Tirz dose from 2.5 to 5mg 3 weeks ago, but I just wasn't feeling it; I figured I probably had to go up even higher to experience more of what that 2.5mg gave me, until I woke up on week 3 and I couldn't even finish a small bowl of oatmeal. This thing hit me like a TRUCK (and gave me runny shits for 3 days), and it all happened overnight. In fact, just a couple of nights before it felt like I was actually getting hungrier.

These drugs take time (sometimes, I felt my initial dose in under 1 hour), and they can be pretty stealthy...

Yeah, that's why it's a bad idea to change dose week to week. At least two, but four is better since that's when blood levels stabilize after a dose change.
 
Somewhere in this thread I've read that the ideal reconstitution leads to .5ml weekly and to do the math around that. I believe it was a post from @Ghoul. Also, I've taken sema before for about 6 months but am now giving Tirz a shot, is it safe to start at 5mg weekly? Thank you
 
Wrapping up the first week on 10mg Tirz. Weirdly enough, it feels smoother than 7.5mg - back then I was wiped out and stuffing food down like it was a chore. Now? Eating feels easy… almost too easy. Confidently chewing through life.

Gonna ride this out for 3 more weeks, and then… well, 12.5mg is just sitting there looking at me like, “you up?”
 
15mg is the best dose I've been on so far. This is my third week, and the gap gets smaller and smaller with hunger. The first week after the bump, I started to get hungry against starting that Thursday afternoon, with injections Sunday morning. The following, second, week, it lasted until Friday. Today, my third week at 15mg, there is no sign of fading. I'd like it to fade a little, as Saturday nights are my once-a-week indulgence. I need to lose about 10 pounds of fat to put me at 6-8% (actually based on last scan, that'd be low 6's). Diet is on point. Muscle loss is minimal. In fact, it climbed a small amount last month.

That's what I'm hoping, but I'm just some collagen-challenged, 52 year old. I didn't start out obese, but there's still enough loose skin that it bugs me. But this last ten pounds is proving difficult. The response is becoming asymptotic. The scale isn't a really good indicator right now because I have found a good recomp window and am gaining small amounts of muscle. But I have veins creeping into view from the perimeter of my stomach and also my lower quads. I use vascularity now more than the scale.

2.50mg is "safer" in terms of avoiding unpleasant GI sides if you're one of the rare hyper responders.

But the majority of males don't feel anything at 2.5mg in my experience.

For the first 4 weeks blood serum concentrations are still rising with the higher dose. Once it levels off, the gap will slowly return and be able to eat more the last couple of days.

I feel like I’m one on the lucky ones. I have been able to stick at 2.5mg of pharma Tirz so far and really feel the appetite suppression effects. I have done 3months in total (1 month with a break of 3 months then 2 months consecutive). Down 4kg (79-75kg) in a recomp now - so stretching out the doses so I can eat somewhere close to maintenance.

I think I noticed the effect in the first week (placebo?), however my advice is to not push the dose too much too quickly

I tried injecting QSC GH IM vs in subq.

Subq I never felt anything or had sides at all. IM, my feet / hands swell, I get the water retention, etc.

Any significance of this? Or just statistical noise?

Sounds to me like IM acted, effectively, like a larger dose of GH.

Since immune response neutralizes the peptide/protein, and it's much weaker in muscle than sub-Q space (which is optimized to deal with frequent infections), maybe that's an indicator you have a strong immune response sub-Q but not IM.

Seems that way. 1mg of QSC unfiltered semaglutide injected into the delt is immediate appetite reduction and gives me a "full" feeling like I just ate a large meal. I'm sub 10% BF, very far below my set point.

1mg QSC sema subq... I don't feel much at all even after X amount of hours.

Yes. It's fine to do it up to 2 days after scheduled dose and return to normal day on the following dose.

If it's more than 2 days late skip it. It has a long half life so you won't fall far behind if you've been using it for at least a month already.

Totally fine. I actually had to skip a full week of Tirz twice last year while traveling abroad - couldn’t bring it with me, so I just did my dose right before leaving (was on 4mg at the time) and picked it back up when I got home. Didn’t notice any difference in how it worked. So if you miss a day or even a few - it’s really no big deal.
 
Am I doomed?

I have been taking Tirz inconsistently for the past months. My work constantly assigns me to travel to underprivileged jungle communities with no electricity - cannot bring my meds.

- November 16 (new vial) - 2.5mg - ⭐
- November 25 - 2.5mg - ⭐
- December 25 (new vial) - 2.5mg - ⭐
- January 2 - 2.5mg - ⭐
- February 15 (new vial) - 2.5mg - ❌
- February 22 - 2.5mg - ❌
- March 13 (new vial) - 2.5mg - ❌
- March 19 - 2.5mg - ❌

The days I marked with stars are the injections that I felt lots of appetite and food noise suppression, literally for the whole week!!! But when February came, it just stopped working for me. I'm really worried that I blew my chances as a good responder and what if I built up some immunity to tirz.

This April they finally found a replacement for me to do the field work, so I'll be able to do Tirz more consistently.
