Giant Semaglutide Thread (and other GLP-1 / GIP agonists)

Am I doomed?

I have been taking Tirz inconsistently for the past months. My work constantly assigns me to travel to underprivileged jungle communities with no electricity - cannot bring my meds.

- November 16 (new vial) - 2.5mg - ⭐
- November 25 - 2.5mg - ⭐
- December 25 (new vial) - 2.5mg - ⭐
- January 2 - 2.5mg - ⭐
- February 15 (new vial) - 2.5mg - ❌
- February 22 - 2.5mg - ❌
- March 13 (new vial) - 2.5mg - ❌
- March 19 - 2.5mg - ❌

The days I marked with stars are the injections that I felt lots of appetite and food noise suppression, literally for the whole week!!! But when February came, it just stopped working for me. I'm really worried that I blew my chances as a good responder and what if I built up some immunity to tirz.

This April they finally found a replacement for me to do the field work, so I'll be able to do Tirz more consistently.
If you’re going to have cold storage problems, could you just purchase small vials and reconstitute them weekly as a single-use thing? I realize it seems wasteful to throw out half of a 5mg or 10mg vial if you didn’t use the whole thing, but it’s not as if it’s outrageously expensive. Obviously you’ve gotta manage your storage conditions a bit still, jungle communities seems like it could be a problem, but the lyophilized peps hold up fine at room temp. Not sure how things go at higher temperatures.

I’d wager a guess your doses are low and inconsistent so you aren’t really building up any concentration of the drug. A lot of folks don’t get any effect at the levels you’re at.

When orforglipron finally comes to market you’ll have a good jungle-ready solution. Not a peptide so should be easy to handle.
 
30mg Sema? How much BAC were you able to get in that vial to reconstitute?

The filters were doing their job, they got clogged by aggregates from what was probobly an insanely over concentrated solution.

What you could have done was transfer to a larger vial, add more BAC, and left it overnight, then filtered it. When you dilute a peptide that's formed aggregates, some may be reversible and can separate back into monomers when the concentration is reduced.

After 3 weeks and using about 10mg sema of the very cloudy Chinese semaglutide, I decided to filter and use more.

This time it passed through the chinese .22mm PES filter easily and was able to filter 9ml without too much trouble. I stopped short of clogging the filter becauase I always wanted to have a fresh filtered batch.

Data point for you. Simply waiting 3 weeks made it much easier to filter. Why? Perhaps you can give your theory.

Aggregates adsorb (stick) to container walls more easily than monomers because they have a much larger surface area to interact with the glass. Since they're stuck in the vial that probably lowered the amount of them in solution so there weren't as many to clog the filter.

When they're really large you can see little spots of "gel" on the vial walls.
 
Wrapping up week 2 of 4 on 10mg Tirz. Last week I was using SSA, this week it’s GA - honestly, didn’t notice much of a difference between the two.

What’s interesting is that 10mg actually feels weaker than 7.5mg did in some ways. But overall, I feel better - no post-injection fatigue like before, and I’ve had no problem hitting my calorie targets.

I’ll keep tracking how things go. Next step: 12.5mg, then 15mg. After that, I might switch to Reta or even try stacking Reta with Tirz. Not sure if that’s the smartest idea, but hey… we’ll find out.

Yeah, but where’s the chaos in that? Just hitting 15mg, feeling stable, and cruising into long-term health like a responsible adult? Sounds… peaceful. Terrifying. I need at least a little self-experimentation and mild panic to keep things interesting!

Has anyone else felt extreme soreness after tirzepatide?

I had been on semaglutide over a year ago. Eventually switched to tirzepatide and did well on it. Recently though I was doing mazdutide to see what that was like. Since QSC is gone I can’t find anywhere to stick with maz. I decided to go back to tizepatide . I started low at 2.5mg injected yesterday. Today I feel like I got hit by a truck. I am sore absolutely everywhere. It hurts to touch my body. It doesn’t feel like aches from an illness. It feels like I did the biggest work out of my life and all my muscles are screaming. I didn’t have this side effect at all with anything I’ve done before.

It’s not a deal breaker, but im wondering who else experienced this.

Your tirzepatide or injection water is contaminated. Your body is fighting off the contamination, which is why you're feeling weak. If it's only mild, you'll experience muscle weakness, joint pain, maybe a headache, or a slight fever. If it's severe, you'll have to spend a lot of time on the toilet over the next few days.
 
Remember, no matter how big the bottle of bacteriostatic water is, it should be thrown in the trash no later than 30 days after first use.

Yes, that's true. I mix BA myself. A 100ml mixture of BA costs me €1.89. Every time I mix a new vial of semaglutide, I make a new 100ml of BA. Otherwise, it would have been eight weeks, and I'd have some concerns. I don't think much will happen even after eight weeks, although countless tests show that most refrigerators in private households are dirtier than toilets. At least microbiologically.
 
It’s gotten awfully quiet around here - either everyone hit their goal weight or decided GLP-1 is yesterday’s news.

Just checking in: I’m wrapping up week 3 on 10mg of Tirz, and honestly, I think I found my sweet spot. No struggles with calories, bloodwork looks perfect, no fatigue… everything just clicking.

Still planning to push it up to 15mg though, because why not? Curiosity is undefeated.

Honestly, Tirz is easily the best thing I’ve ever used. Without question, it’s taken the #2 spot in my book - right after good old testosterone.

Thye're not as "novel" anymore; we all know they work, we all use them now. I don't think we've had something this "magical/ground breaking" in a very, very long time.
 
what you guys think of cagrilitide? I haven't looked into it much but wanted to ask some opinions on here. Do you think it's a worthwhile addition to sema or tirz?

I heard people say it works more by mentally making you less inclined to eat rather than phisically suppressing your appetite? is that true? if so it would be very usefull to generally reduce craving without making you unable to eat. with tirz for a while I had zero inclination to eat but I was eating so little it wasn't good for muscle. Now that I lost weigt is stable the appetite is back but thinking about food all the time sucks. I will increase my tirz but I think the same will happen, I'll get a bit leaner and then the fold noise comes back, maybe cagrilitide could help that? I don''t even want to loose too much more weight, like 5-8lbs at most. I just wanna eliminate the food noise so I can be in a better mood

Yeah, every Sunday like clockwork. Sometimes if something came up, I pushed it to day 8 or 9, but mostly it’s every Sunday.

I’ve always had pretty good control over my appetite, even before starting, so I never really had issues with crashes, food noise, or crazy hunger showing up around day 5–6 like some others mention. It’s been smooth for me all the way.
