Giant Semaglutide Thread (and other GLP-1 / GIP agonists)

How many weeks did you run the 15mg Tirzepatide dose? :0 Also, how long do you use a bottle of bac water before opening a fresh one?
only 1 week on 12.5 and 1 week on 15. but always I always felt the jumps right away. from 2.5 to 5 and from 5 to 10 worked instantly.
I'm not sure about the BAC, definetly a while but it's the same as I always did with all of my vials and never had any issues. I just use it until it's finished. maybe 2 months
 
Anyone might know why more than 10mg Tirz seems to not result in any more effect for me? I have tried 12.5 and 15mg with no difference at all from 10mg.

because you are below 10% bodyfat and have slowed your metabolism by starving yourself in addition to the appetite supression from tirz

you dont know anything about dieting and you just think its best to eat less and take more GLP, it is not this simple at non obese bodyfat %

You are insanely lean already. With all the restrictions you’ve tried, it seems like your body is saying enough. Not trying to pile on, but other posters might find it helpful to see what you actually look like when wanting to get even smaller.
 
So it’s been a week & some days. Did my 2nd injection on Monday

So far… it’s been honestly nice & no noticeable sides. My main thing I’ve had to deal with is my insulin, immediately after my first injection I could tell my insulin sensitivity increased & in my case that’s pretty good & a sign my Tirz isn’t bunk. I’m injecting a good 30%-50% less insulin for the same amount of carbs & I’m pretty happy about that part. The issue comes in with Tirzepatide & its digestion slowing attributes.

Initially my sugars are better but it isn’t until a handful hours to a whole day afterwards & then some that my sugars seems to be much more difficult to bring back down. I’m needing to take higher correction doses more frequently, in my case Humalog since my sugars start to spike pretty quickly but then I get nervous of Insulin stacking & how I might go too low right before bed. Outside of that my sugars have been stable if a little higher around the 150s-170s

Not hard to deal with either as I’ll eventually figure out a new insulin regime but hopefully it becomes easier to deal with as my body gets used to Tirz.

That being said I haven’t paid much attention nor worked on my weight. I’ve been more focused on what & how much I can eat while on this medication. I’ll get back to schedule next injection but the suppressed food noise is pretty cool, I can go all day & legit forget to eat if i get real busy, that & the taste of food seems to have changed a bit. Most sweets are way too sweet to enjoy lol, can barely get a few bites into some before having to put them away. There’s me being full before finishing my plate too. Been leaving a good portion of my meals for later on or the next day.

Oh and cans of sodas (diet/zero) are much harder to finish just something small I noticed, other than that, no sleep issues but I can’t say about elevated heart rates. I should monitor that later
 
I dropped my dose to 6mg of tirz, might end up going back down to 5mg. 7.5mg just slowed me down so much i was getting heart burn and the digestion was affecting my sleep. two cups of cottage cheese before bed was messing me up. i think ill just do plain chicken breastnow

I got this garmin watch to track my sleep, and even with wearing cpap for a few hours, melatonin, glycine, magnesium etc, my sleep has been crap. lots of tossing and turning and 'light' sleep.

sure Im running tren but i really think its gut related, I don't have much of an issues falling asleep or staying asleep we'll see.

like i've posted before, my hba1c went from 5.3 to 4.9 while gaining weight, while going from 1mg slowly up to 6mg, so I don't think i need to be at 7.5mg+ unless i needed appetite supression.
 
Reta finally feels like it’s kicking in. Either the buildup effect has started or my body has adapted, but this last week on 4 mg has been different. The sugar cravings are gone, my energy is stable, and none of that sudden fatigue I always had on Tirz. Next week I’m thinking about increasing to 5 mg instead of jumping straight to 6 and seeing how it goes from there.

So far I’m happy with it. Appetite is solid, hitting calories is easy, but the “switch” in my head always turns on when I’m about to overeat. No regrets about switching from Tirz to Reta.

As a rule. It takes 4 weeks to reach stable plasma levels with GLPs, so max impact of a given dose isn’t reached until that point.
 
Nah i mean i get it and seems like its a fact since its writen everywhere just i personally never noticed diference except moving up a dose each 4weeks felt the same or weaker

I mean pharmacokinetics are a fact, it’s based on the half life of the drug. Everyone’s metabolism is different of course, so it might take less or more time to reach stable levels.

But going up a dose and it feeling weaker is weird. It sounds like immunogenicity (your immune system neutralizing the drug).

Thats something unappreciated about UGL, and one of the reasons to filter (to remove aggregates, a primary cause of developing immunity to the drug, more than once a week injections are another). Jano recently upgraded his equipment to test for GLP aggregates because he’s had requests to check for it.

Pharma doesn’t have this problem with aggregates, since they focus on preventing it. The FDA routinely buys peptide/protein drugs “off the shelf” from pharmacies to make sure aggregates don’t form as the result of uncontrolled temps in storage/transport.

Unlike our UGL kits that could be sitting for weeks in a box at the top of a Chinese storage container where it hits 150° f+ in summer.

After this letter came out, the FDA did some testing of cheap compounded Semaglutide, found the immunogenic response approx 2000x worse than pharma. Filtering it lowered that by 95%.
