Help brewing L carnitine

I make pipless L-carnitine 500mg/ml regularly, from bulk supplements.

264 ml distilled water ( heated /mag stirrer)
5ml HCI
Slowly add 250g L-carnitine bulk sups
( let stir for a few minutes will be a lil cloudy)
Add 5 ml BA
Add distilled water until you hit 500ml staring the entire time.
Filter through 22um filter (I use buchnar funnel with medium filter its actually a 20um) comes out crystal clear and ph is around 6-7
If you want to stick to something similar to the funnel idea but actually safe and sterile

Use a stainless steel clasp style

Buy 5 of them or more. They are great. Will never break. The filter is clasped using a special gasket and stainless steel plate. Zero bypass risk. Can be autoclaved. I use autoclave bags. Can put filter inside and just go to town. Can stack a pre filter on top if needed.

Accepts 47mm filter paper is readily available

PTFE is my preferred choice but I also use PES when I want water based.


Throughput is high and if your able to be quick and aspeptic you can hot swap filter paper but I tend to just swap funnels instead. Safer. Buy a few. They are very useful.


Your bottle top filter is fine however for water based. But if you do steroids I really recommend this. Many benefits going this route. Did I mention fully 316L stainless across the entire filter path?

 
To make the L carnintine more isotonic I’ve read NaCL (sodium chloride) is very effective
Or you could use Glycine

My magnetic mixer comes this week and I’m going to give this recipe a try unless any of you have strong opinions about this recipe being bad.

-Lcarnintine base 25g
-Taurine 5g
-Sodium chloride .45g (help make it more isotonic)
-sodium citrate .10g (ph and buffer agent)
-citric acid .005g (ph and buffer agent)
-Benzyl alcohol .75ml
-sterile water 35ml initially just for displacement purposes then add remaining to get to 50ml

50ml multi use vial
500mg/ml L carnintine
100mg/ml taurine
Aiming for a ph level between 6.5 and 7

This will be my first time making L carnintine so we will see how it goes.
Sorry I forgot about this thread for a minute and might be way behind but adding NaCl with increase the tonicity even further. It increases the number of ionized molecules

At 500 mg/ml, L carn is already extremely hypertonic.


500 mg/ml is about 3.1 mol per liter, which works out to over 3000 mOsm per liter. Normal plasma is about 280 to 300 mOsm per liter. Tonicity depends on the number of dissolved particles, not their weight.

When sodium chloride is added, it dissociates into Na+ and Cl- ions, creating two osmotic particles per molecule. This means sodium chloride adds osmoles and does reduce tonicity.

Because tonicity is additive, the osmotic load from L carn remains and the nacl increases it further. The result is an even more hypertonic solution, which pulls water out of tissue

Even though people been doing it for years it’s outside the safety margins of any IM injectable. (I dilute mine)

How much do you dilute?
 
When using distilled water is this boiled and cooled first before heating back up to add BA or is filtering at the end enough
