HMG reliable analysis

Hi,

Am using HMG from SSA for a short while. Saw their test from Janoshik with 7,5mcg. I found out that is it pretty difficult to translate HMG from IU to mcg. Another analysis from QSC HMG last year was 214mcg ...Janoshik answered me on this topic that he cannot translate his result in mcg to IU because he needed data from the Factory...
Anyone an idea of this?
 
If I had it, I'd do so.

Maybe ask Chromate, he's delivering results in IU right away, is he not? :)
 
IU is a measure of bioactivity (potency).

Unless a lab performs a bioassay, any IU provided is just an educated guess, based on assumptions. There are factors besides chemical purity and mass that impact how well (or if at all) molecules attach to receptors, so 1mg of two different batches of the same substance, at the same purity (as measured by HPLC, and sometimes even MS) can have different potencies (IUs / mg).
 
I was considering an FSH ELISA to screen a few different HMG products that I have on hand. But, I am bogged down with other projects at work right now. For the moment I am going use some menopause urine tests to at least confirm that FSH is actually in the products. Not many vendors carry HMG to begin with, and on top of that, there is rarely any form of testing. So, it ends up being a leap of faith as to whether it is even HMG, which frustrates me.

Right, but there must be a general mass range of HMG in let's say 75iu preparations. We could then assume (big assume) that the 250mcg contents is roughly...10iu...200iu, ECT. Yes the bioactivity is a big guess, but is it anywhere near the expected mass ballpark.
 

Sorry man, bioactivity testing can be done, but it's very expensive. That's why the info from the manufacturer is needed. They'll have the "IU per mg" data from their own bioactivity testing which can be used to translate purity tests from Jano into the actual IUs of activity.

That study makes the observation of improvements in total non-bioactive protein dropping over the years across better types of HMG, as well as mean specific activity (IU/mg) across types. Seems like while an ELISA test would be very exact, there is some general ballpark of IU/mg relationship per type of HMG.

Along this line, you have tons of data. Any clue as to the rough mean or median in mcg of HMG you have tested for 75iu preparations? 150iu is pretty rare.

Is it safe to assume that Chinese/non-pharma is recombinant preparations and not urine purified?
 
It's complicated, isn't it? HMG is not a hormone like hCG. HMG is a mixture of two hormones, LH and FSH. So, I would guess (but don't know) that if something is claiming to be 75 IU, it contains 75 IU of LH and 75 IU of FSH.

I would assume most people who are interested in HMG are already taking hCG. Since the hCG is already mimicking LH, the primary desired hormone from HMG would be FSH.

FWIW, I did test a sample of HMG with a menopause urine test and it tested positive. Far cry from a quantitative measurement, but at least it confirms there is FSH in the vial.
