Transfering compound from media bottle

I have used both syringe filter and bottle top to media with a septa cap into sealed sterile vials.

I am looking to cap n crimp my next brew. The method I feel would be best is sterile pipettes and a pipette dispenser (SEOH on Amazon).

I am a little worried about the top on the media bottle being exposed.

Any feedback, suggestions, or techniques would be greatly appreciated! Thanks guys.
 
I use a pipette and I've never had and issue . The main thing is sterilizing your work space . Wipe down all the work surfaces , the walls , sweep the floors , ECT. The more thorough you are the better.

The other thing I do is cover all the vents in the room that I brew in to cut down the airflow , so you don't potentially kick up a bunch of dander.

Even with the septa' s your open vials are still exposed for some period of time. So even if you use that method your still gonna get some exposure and it's a lot more tedious and time consuming than using a pipette. To me it's just better to go over board on sterilization then transfer the gear and cap it as quick as possible.

Appreciate the feedback. Yeah I was thinking of getting some plastic (perhaps mattress bags) and make an enclosed workspace.

I didn't plan to use septa caps with unsealed vials. Seems like the tedious aspect of it leaves too much exposure time for the vials. Which the open media bottle kinda has me worried that the brew is exposed to air. (I guess it's not that bad if the environment is contained?)

Do you think sterilising the vials with the foil on top is the way to go? And just poke the pipette through it?

I still sterilize vials like normal and put Tin foil on each vial and bake them , but I don't poke the pipette through the tin foil since it isn't a sterile surface . I take the tin foil off 10 or so vials at a time , pipe my oil into a vial and put the stopper on and sit it off to the side . After i work through those 10 vials I throw a cap on the media bottle, prep another 10 vials and rinse and repeat the process until I work through my way through all of my oil . I at least try to cut down on the amount of time my oil is exposed , but if you use a septa or pipette your still gonna have some amount of time that it's exposed to air .

I've seen that they make these clear lab bags that are similar to a sterile filling station with built in glove holes , never personally tried them but it's something worth looking into if you want to go the extra mile for sterilization.
 
I filter into a media bottle and then use a spin needle which is like 6inches long and I'll crack open the filter just enough to slide the needle in and draw out. When I'm done drawing up I slide the top back on and I attach a 25g needle to my 60ml and use that to dispense into pre-sealed vials.
If I don't use pre-sealed vials then I wait till I'm ready to fill and I'll pull them out hot and fill them and put a stopper on right away. This action is done very quickly.
Everything is inspected before and after. It takes up a lot of time for me so I tend to steer away from this. Plus I'm over paranoid and I hate the anxiety.

If I do small batches I use a syringe filter cause there's nothing to clean up or prep. I just set up a device to push down on it all the time so you don't have to keep squeezing on a calling gun.

Yeah guys I appreciate it. Just trying to stay safe. @LordSamuilo do you use a fresh pipette every time? The question sounds kinda strange in my head. Lol
 
Pipettesdotcom has electric pipette device you can sit on your media bottle. Not sure if they're sterile but I'm sure if you start there you may find something that can help you. I don't see why they can't come sterile. They have one that's digital for $800 but I saw some as cheap as $100.00.

I just went ahead and ordered the 96 well bench top pipettor. It was only $16K.... :D:eek:

There is a ton on that site to look at. It seems a positive displacement dispenser is what is needed. I think the Amazon one will do the trick. That bottle top pump dispenser for 100 would be real nice. But no autoclave to reuse it.

I'm beginning to think I'm over thinking this. Lol

Lmao , shit you couldnt order me one while you where at it , thats fucked up man ! :D

I finally coughed up the money and bought a pipettor recently (200ish $ not 16k:oops:) but before that i just used single use disposable pipettes off Amazon. Theres a shit ton of different lengths and volumes , youll have to search a little to find what you need but they're dirt cheap.. definitely the most economically friendly option.

Haha yeah I think the 10$ one is good enough. Unless Oprah wants to pass out some pipettors.

The sterile pipettes that go with it are 10ml and 10 for 5 bucks. I'm only gonna brew 450ml test E and a lil deca to finish this blast. But I want to hold onto these vials for a bit. Hence going the capping route.

I use a lot of syringe filters and brew small amounts, so I typically filter into a 100ml vial with caulk gun setup then rip the aluminum seal off and pop the stopper off and pour into 10ml vials. It works and I've never had an issue. If I was going to brew larger quantities a 10ml pump is the way to go.. just pump and silicone stopper then go back and aluminum seal them all.

Both methods require clean room type environment to ensure sterility.

Your not over thinking just throwing out ideas till one sticks. Someone mentions a 10$ pipette and someone mentions a 16k dollar one and what you need is somewhere in the middle. Who knows you might end up spending a couple hundred instead of $50 cause it fits your needs better and most of all makes the process safer.
I'm not kidding when I say that I've come up with some of the best ideas cause me and few friends were just spewing out what came to mind.
You said no autoclave? Well, I guess I need to make an autoclave at home. Lol
 
True indeed. I ordered the SEOH pipettes and dispenser just to play with it and see. They're too cheap not to mess with. It might just work out!

One other thing I noticed last time I brewed was my hose on my vacuum pump was collapsing a decent amount. Is this normal? I let it run through the whole process but felt like I should have turned it off every now and then.

Most dispensers can't be properly sterilized without an autoclave

At least use a pipette + pipette pump

At least the thing touching the liquid (the pipette) is made of glass and can be sterilized with alcohol, hydrogen peroxide, some even baked (make sure they're made of heat resistant glass)
Also, some pipette pumps withstand alcohol.
 
I sterilize my vials and stoppers separately and then lay the stopper on top halfway in the vial (seals it but its not completely seated in the vial) and give it some more heat. Then I only lift up the stopper long enough to fill the vial, less exposure to air that way.

And I use a 20ml sterile syringe with a 18g 1.5in needle on the end to pull out of media bottle and put into vial. The 18g is big enough that it doesn't restrict the flow of oil too much and the big barrel and needle are cheap and pre-sterilized...

And deff turn off the all fans prior to transferring. No floating stuff in the air!!!

I just got the pipettes in the mail. They come individually sealed and sterile. I didn't expect them to be ten inches long. Lol. But the dispenser is on the way and it looks exactly like your picture

Any chance you could elaborate on this in a little more detail? I'll have to look up what a spin needle is. I'm so done with the fucking syringe filters lol

A spin needle is just a really long needle. Sometimes it's 4-6 inches to help get to the bottle of the media bottle after slightly opening the top.
