hey guys quick question regarding open vial sterilization.

an Isopropyl alcohol bath followed by covering with aluminium foil and bake is generally the quick dry sterilization procedure.

My question is: before covering with aluminium foil do I need to make sure absolutely all the IPA has evaporated?
If so, technically couldn't contaminants enter the vial at this point also?
 
Isopropyl will sterilize everything it touches quickly. The heat from an oven should expedite the evaporation of the alcohol if allowed. If you cover them with foil you'll create an alcohol steam bath. Either let it all evaporate first if you're going to cover it or pop it the oven wet and uncovered. The heat will do the rest.
Contamination is always an issue. Dust particulates can be visible in the oil. You just have to be careful. No kicking up dust in the kitchen.

The alcohol will STERILIZE nothing, heat of vaporization of the alcohol will not in any way shape or form make an alcohol steam bath. You may disinfect but IsoP will do little else. Without direct rubbing the alcohol will not penetrate microbial cell walls. Before you answer a question do a little research, false responses are much worse than no response. Heat will sterilize nothing you may get close with 2 hours at 170 Celsius. The ONLY way to sterilize anything is with an autoclave but heat sterilization will get you close. I have sources for this information but I am too lazy to post.

Okay. I do know that the alcohol penetrates microbial walls through osmosis, denaturing/ cooking the microbe, then evaporates taking the cooked nucleus with It. I also know that it's not necessary to rub isop on a non-porous surface to sterilize it. You may spread it but rubbing is't necessary.
I do agree It doesn't kill everything.That was a neglectful statement above. I don't give much thought to viruses given our bodies manage most of them well. I'll manage my Ps and Qs in the future. I should have just suggested an autoclave.

Also in response to yours. If alcohol evaporates then has no where to go it will condense when cooled. What I was suggesting is not to cover in a way that won't allow the alcohol to evaporate into the atmosphere.

Jesus, if you have a formal education that includes microbiology you schools failed you. Microbial walls consist of amino sugars known as peptidoglycan, alcohol does not use osmosis(alcohol is not water) to cross the membrane it uses diffusion. It is necessary to rub alcohol on, why do you think when you are given a shot you get alcohol rubbed onto you? Without the rubbing you do not adequately break the murien(peptidogylcan) walls.
Where the fuck did you get that the nucleus cooks and evaporates out of a cell? Bacteria do not even have a nucleus! They have genomic code scattered about and some may be concentrated in areas this would be the nucleolus. Alcohol does not denature any proteins but heat does. The problem is endospores, which can survive extreme temperatures in excess of 1500C.
Why did you bring viruses up? Nobody else did and your previous statement included enough microbiological ignorance you could have stopped there. Yeah I dont worry about viruses because my immune system beats all of them, Ebola, HIV I don't worry about any of that nonsense. Why even bring up viruses?!
Your suggestion to put everything into a sealed container and cook it will yield little more than a bomb.

So what is the bottom line. Are you saying 170 degrees C will sterilize a vial? How long must you heat the vial, i'm kinda confused. What is a method to sterilize a vial without and autoclave?

Okay, just my bro knowledge and hopefully someone with some additional education can chime in...but my 2cents...Lets say you dont have an autoclave and youre going for the "best sterilization" you can do, my guess would be you would best served by doing a hydrogen peroxide bath, then bake at 170 for a couple hours. While this might not kill everything it should wipe out the majority of bugs and a healthy body should handle the rest.

Maybe we should point this question in a specific direction.

"If you dont have an autoclave what is the best "sterilization" technique?

Cooking the fucking nucleus off?! Where some of you people get these fucking ideas I will never fucking know. And when somebody calls B.S. because you clearly didn't do any god-fucking-damn research and you have no education on the matter (WHICH YOU KNOW PERFECTLY WELL), you turn around and offer MORE ridiculous shit? There are people around who really DO KNOW what the hell is going on, so let them answer these questions. This is important stuff.

Wouldnt hydrogen peroxide work better than alcohol in this scenario?

I know they use HP gas in low pressure autoclaves for sterilization. With claims of sterilization in 6 mins. Again, Im just kind of taking bits of things and putting it together for a somewhat logical guess. :D

Obviously an autoclave is best but it appears most dont have access to one so it would be nice to have a second best option if there is one that can provide safe results.

There are large amounts of studies on 170 Celsius heat sterilization, we use it in my lab on somethings and I am comfortable saying I would trust it. Hydrogen peroxide would probably not work that well there are several genus' of bacteria that use H202 to break down reactive oxygen.

What I did learn and didn't know is that alcohol doesn't kill bacterial spores along with protozoa and nonenveloped viruses. Protozoa are not so common unless you're swimming in dirty water. As you may assume there isn't a lot of aids and hep c splattered around most domestic US homes. So If you'd like to worry aids and aliens and shit that that's cool with me but it's a poor argument.
So crack an egg in a bowl. Poor rubbing alcohol on it and wait. You can scramble it to speed it up but what happens in the alcohol denatures the proteins which is what you do when you cook food. Bacteria remains small enough to then be carried off in the vapor.

Where the fuck are you coming up with this shit? Vapor alcohol is in no way shape or form picking up microbial cells the vapors have no where near the kinetic energy to do something like that. Not to mention they do not have hands and are not picking them up. You are a stupid person in this area, stop propagating your lack of knowledge on these matters.
An average microbial cell is at least 2.3 microns in diameter and you think molecular alcohol picks these up!? I laugh at your ignorance, literally you are the making me laugh. I will share your lack of knowledge with others so they may have the same entertainment as I.

I rinse my media bottles with alconox to chemically clean them then I do a 5 minute rinse of ultrapure water (you do not have access to this so DI will do) I do this 3 times. Then without wiping I send them to the autoclave or in your case the oven.
