Most interested in cagri, a different pathway from the glp1 gip glucagon approach. Should be cool if you could come off your tirz/reta for a month, throw in cagri for 30 days, drop your tirz/reta tolerance way down and start back up after the 30 days with a fresh tolerance to save money, would never have to really get up to significant doses of either. I think stacking it might be most people's interest though. Lots of stuff of the horizon!

Exactly, it might even be healthier in the long run too. As opposed to just stimulating the same receptor(s) non-stop.

When do you bulk for muscle in all this?

It is not super hard to get in excess calories on low to moderate dose tirz. I would argue being super meticulous about adhering to a weight gain diet that has everything weighed and measured for .25-.5% bodyweight increase per week will be more productive than a more food more better bulk. @Spaceman Spiff has a thread about his 5th cycle and used tirz at 5mg on a bulk and I believe he saw it as useful addition.

These GLP/GIP/Glucagon agonists also have the added benefit of increasing insulin sensitivity and release, both being helpful when in a slight caloric surplus.

sema is definitely not for me, I have gone up to 2.5 and I don't feel anything, maybe the first 2-3 days a small reduction in hunger, I don't know if I should try 3mg as the last dose and if it doesn't work change to another LPG

No way, i already have an extremely hard time getting in the cals needed to bulk on my size. I can eat no food for a day or two easy. So will not utilize. Odd

Have you looked into this one, or the MOA? Couldn't find much but fatty diarrhea or nausea/bloody stools in mice? I think they tried MGAT inhibitors before and it didn't go well. Maybe S-309309 is different.

Yeah, I couldn't find much about the MOA either but in one of the references on Wikipedia it does state: "MGAT2 Inhibition Activates NTS Neurons, Leading to Appetite Suppression and Beneficial Effects"

I can't find that particular study though, it might be listed somewhere in a different language, but I have no reason to believe that the title is made up or anything like that.

There were also 2 interviews with the company behind it. In the first interview seen here, one of the questions is if they are considering making it a combo treatment with another drug. And they replied saying they are considering it and that it has "great potential for combination administration".

In the second interview this part stood out to me:

"It is likely to involve a wide range of treatments, including not just monotherapy but also maintenance therapy with various concomitant drugs"

It does make sense because the weight loss is less extreme than other medications, as the rep points out here:

"8% to 10% weight loss range can be achieve within 24 weeks."

They also mention that it will be an oral drug. Thanks for those links by the way, that's very interesting.

Yup, Type-II has a good article on it.

Buyer beware: @lift4lyfe I found it difficult even eat at maintenance on Sema, let alone slight surplus.

Overall it was net zero gains for me. Unless you're new to weight training and start it the same time you start GLP1s, probably gonna lose muscle w/o PEDs.

Three more, I will keep updating as more are coming out:

Ecnoglutide
cAMP-biased glucagon-like peptide-1 (GLP-1) analog
Code:
https://pubmed.ncbi.nlm.nih.gov/37364710/

Survodutide
glucagon/GLP-1 receptor dual agonist
Code:

Pemvidutide
dual GLP-1/glucagon receptor agonist
Code:
https://en.wikipedia.org/wiki/Pemvidutide

Agreed. Most people who are obese, this is not a problem, and the reality is we are not going to lose the massive weight required to get to a healthy size without some muscle loss under any circumstance.

Very much want to hear more about what the Meso minded people are doing to keep muscle while on these, I'm using GRF 1-29 to try and help, but I'm still so hopelessly out of shape that I don't feel the need to do anything more. That will likely change come the fall.

Original plan was to swap from Tirz to Sema for for a long period to maintain my set weight before titrating off altogether way down the line, but Tirz has gotten so cheap I won't have to now. Love this idea of switching to cagri for a long time if it can be shown to be safer.

I stopped using tirz in December. Got back on in April. Trying to get lean. Isn’t helping with those last few pounds which I guess it’s. It really for that anyway. Got some mazdutide and cagritirz to try in a few months. But my weight training goes to crap when I am on glp. Kind of sucks.
