Giant Semaglutide Thread (and other GLP-1 / GIP agonists)

Came across this and thought it was kind of cool so wanted to share. Plots out your dose and half life to see how long it stays in your system. Was really interesting to plot T and R and see how much longer R stays in your system. Just a nice visual.

 
Just a PSA about this calculator from our friends on Reddit. Semaglutide induces drug neutralizing immunogenicity. Once a week dosing kept this immunity from developing beyond a minor level.

When 95%+ of pharma protocol Sema and Tirz users achieve significant, durable weight loss with no loss in effectiveness for years, but the Reddit scientists find every compound "stops working" and they end up stacking Sema/Tirz and Reta, using daily micro dosing, yet 300 injections later can't seem to lose the 10 pounds they've been trying to for the last year, consider this may be the problem.

I was not thinking about the micro dosing part of it, it was how each one stays in your system so differently. Just a visual for half life and how they build in your system.

Not a dig at you at all, the more info the better we can make our own choices. I'm actually ok with volunteer guinea pigs, merely pointing out that just because the developers of these drugs can fit the instructions for use in two sentences on the box of pens, doesn't mean they weren't developed by carefully evaluating hundreds of factors over the course of years, immunogenicity being one of the big ones.

99% of users don't even think the ratio of BAC water to dose makes any difference, and choose based on whatever's convenient. #pharmacokinetics #aggregation

I was comparing .75mg R and 4mg Tirz so I guess my comment came from how much slower the .75 was being absorbed vs the 4mg Tirz. Sorry I should have stated that and it only affects me personally so probably shouldn’t have said that at all. Lol

My post should say I was NOT thinking about the micro dosing part. Forgot the not. ‍
 
While I agree, I think the most important factor why they chose the 7 day injection frequency is mere convenience.

That's not to say that I would recommend doing more frequent injections, quite the contrary actually. But I don't think there is harm in doing a bit less, like 5 days with Tirzepatide...

I'm sure that was a factor. But that's where we know, with certainty, immunogenicity is not a problem. Had they conducted testing using a 5 day dosing schedule, perhaps 5% or 10% would've developed a problematic level of immunogenicity though. We just don't know, because that's not what was measured,

If the company said "let's offer a 5 day dosing frequency option in the directions", because it's a protein based drug, the FDA would forbid that until it was proven immunogenicity wasn't a problem at that frequency.

So then the question becomes, why take the risk, however small it may be perceived? Not losing weight fast enough? How impatient can someone be?

Agree completely and actually I liked on both sema or Tirz (more in tirz) that the last two days before the injection I felt hunger coming back or a less slowing of gastric emptiness. It was refreshing.

Ppl that have a problem with that should work on their psyche

The risk for us using this unregulated stuff is actually far higher because they're hardly the "clean" peptides pharma used in their trials. I have little doubt in a few years, we'll read case reports of people who've developed immunity to natural GLP and GIP, and suffer serious metabolic problems, like taking an "anti-GLP".

All the more reason to reduce risk by minimizing frequency of exposure.

I like that feeling too, couple days before I don’t feel the effects at all but gives me a couple days to feel where I’m really at. I look forward to them. ‍‍
 
Sorry but what does immunogenicity mean? and why don't we want it?

It's the immune system's reaction to peptides. An immunogenic reaction creates antibodies that neutralize the drug, making you immune to it, so the drug becomes less, or even completely ineffective.

It's what a vaccine does intentionally, but
in this case unwanted and major area of concern for pharma and the FDA,

Another possibility with peptides that mimic hormones, like GLPs, is developing a cross immunity to the natural hormone, which can be extremely serious,

It can't always be avoided, but minimized by controlling factors like frequency of dose administration, using the correct ratio of water / peptide reconstitution, and preventing the peptide from degrading, which all contribute to worsening immunogenicity risk.

Won't help domesticated dogs lose weight apparently, but would work on cats.
