Use smaller gauge like 25G. You can draw no problem with 25GMostly 22gauge.. sometimes the hole seals up sometimes they don't very well.. since I filter againg with syringe filters when I transfer i don't even use the septas anymore. I try and vial up everything I make.. and if I can't i just put a regular cap on it to store. My process may change in the future.... to gear more twards cutting out syringe filtering all together like you are wanting to do. !
If you transfer from a sterile container to another sterile container using a sterile vehicle between the two I don't see how any contamination can happen.TLDR: If an average of 6% of compounds transferred between containers in a pharmacy ISO clean room introduce microbial contamination there's no way that's not happening in home environment. It's 0% with a hood.
Don't mistake the "I injected it and it didn't kill me" with free from bacteria. Most bacterial infections are asymptomatic, but can go on to damage joints (reactive arthritis) and organs, incrementally, over time.
It's up to the individual and their risk tolerance, but a hood is far from useless for those who want the lowest risk of long term harm from contaminated injectables.
As far as Jano's sterility testing, the focus is on the wrong thing imo. "Sterility" means an environment where bacteria can't reproduce. It doesn't mean your vial, made with non sterile ingredients in a non sterile environment, isn't packed with bacteria, that, once injected are no longer prevented from reproducing by benzyl alcohol. It's your immune system that's the final filter. A LAL test is required to measure the presence of (the worst) microbes (which also measures endotoxin, and combined are called the "bioburden").

If I draw with a sterile syringe from a sterile media bottle and inject into a sterile sealed vial how can it get contaminated? Magic?
No 25 gauge does not draw worth a fuck for filling up a 60 or 100ml syringe.. 22 gauge is really kind of depressing as well.. I have some w 16 gauge that works like a charm.... I just screw the top off use those..Use smaller gauge like 25G. You can draw no problem with 25G
Now I understand better and yeah I completely understand what you are saying.Every time a vial is penetrated in a non sterile environment microbes are introduced. That's why multi-dose vials have to have BA, and ampules don't. Once a sealed ampule is (heat or radiation) sterilized it stays that way.

They're using sterile needles, sterile pharma product, and presumably sterilizing the vial tops yet significant levels of contamination are found in multi-dose vials.
My comment was more about filling open top vials without a hood, which is the worst case scenario, but even needle transfer in open air causes it. That's why Testosterone and other pharma injectables may have a multi-year shelf life, but instruct you to use them within a month of first penetration of the vial.

Again, not everyone cares, nor do they have to. But it's good to know you can't usually "feel" bacteria, that they can hurt you over many years, and what it takes to reduce them, for anyone who wants to go to the next level.
I use a very similar method, except I use 50 ml vials. If at some point in time, it looks like the stopper is getting too used/abused, I just rip off the whole lid and replace it with new rubber stopper etc--it usually doesn't happen though.Paralysis by analysis... BD used to mix oil, solvents and raw and just pour it in open vials, not sterilizing shit in the process. Sold tens of thousands such vials. Put 5% BA and done lol!
Don't over-complicate. Bottle-top to sterilized glass gl45 media bottle. Screw gl45 cap with septa. Use a sterilized glass syringe or a regular sealed one with a long ass needle (sterilize that with your vials and stoppers). Have your vials with the stoppers (no cap) ready.Remove stopper, put 10cc there, put stopper back. When you finish all your vials, cap em and crimp them.
DONE.

But my overthinking brain was thinking isnt it better to turn it off before brewing so the air isnt moving?Yeah indeed, consumer UV lamps can't sterilize far away tho so for example it's better to keep it very close to the place/tabletop you gonna brew, run it for the time it needs and then keep an air purifier on so that the room continue to be as sterile as possible.
Enter the room after the sterilization have everything set up before so that you just start brewing, best is to clean with bleach and then iso before doing everything then final sterilization with UV lamp + air purifier the whole time (or after the UV lamp has finished).
This one way to do it, anorher one could be to sterilize with bleach than iso, start brewing WHEN you are ready to cap the vials and fill, you keep the brew warm and sterilize everything with UV lamp + air purifier and then you cap it. I mean one way or another it's really a lot more then average UGL is probably doing xD
Next step after this is getting a laminar flow hood, you get that one you are set for life. Can't beat that shit, is it necessary? Probably not, but sure as hell I'm gonna buy one when I get my home built, it's too cool ahahha

Best for a medium small room is: Levoit Vital 200s or Winix 5500-2. Price are 150-200$ and it works well.
Keep it at maximum intensity 30-45 minutes before brewing and during the whole time.

That little flow hood is awesome!!!!! I refilter in sterile vials BUT then after pop the cap and put a new stopper and cap. Again overboard but I feel betterYou don't keep it close to your brewing station when you are doing sensible thing so it doesn't really matter, I guess.
You keep the air clean if you keep it running even so it is already clean if you have used it in a closed room for like 1 hour so... I believe on or off doesn't really matter.
Are you capping your own vial? If no you don't need to worry about air moving really.
If you are capping your vial and you wanna go overboard and I have been assured by ppl that have done this for 20+ years IT IS NOT NEEDED, so let it be clear it's just for your own personal peace of mind and being like me autistic 100%
